Whenever I see my 10-year-old daughter brimming over with so much energy that she jumps up in the middle of supper to run around the table, I think to myself, "those young mitochondria."

Mitochondria are our cells' energy dynamos. Descended from bacteria that colonized other cells about 2 billion years, they get flaky as we age. A prominent theory of aging holds that decaying of mitochondria is a key driver of aging. While it's not clear why our mitochondria fade as we age, evidence suggests that it leads to everything from heart failure to neurodegeneration, as well as the complete absence of zipping around the supper table.
Recent research suggests it may be possible to reverse mitochondrial decay with dietary supplements that increase cellular levels of a molecule called NAD (nicotinamide adenine dinucleotide).

NAD is a linchpin of energy metabolism, among other roles, and its diminishing level with age has been implicated in mitochondrial deterioration. 5Linx Edge and Fuel contain nicotinamide riboside, or NR, a precursor to NAD that's found in trace amounts in milk, might be able to boost NAD levels. In support of that
The NAD story took off toward the end of 2013 with a high profile paper by Harvard's David Sinclair and colleagues. Sinclair, recall, achieved fame in the mid-2000s for research on yeast and mice that suggested the red wine ingredient resveratrol mimics anti-aging effects of calorie restriction. This time his lab made headlines by reporting that the mitochondria in muscles of elderly mice were restored to a youthful state after just a week of injections with NMN (nicotinamide mononucleotide), a molecule that naturally occurs in cells and, like NR, boosts levels of NAD.
It should be noted, however, that muscle strength was not improved in the NMN-treated mice—the researchers speculated that one week of treatment wasn't enough to do that despite signs that their age-related mitochondrial deterioration was reversed.
NMN isn't available as a consumer product. But Sinclair's report sparked excitement about NR, which was already on the market as a supplement called 5Linx Edge (Niagen) along with it’s catalyst 5Linx Boost (pterostilbene) .

The NAD findings tie into the ongoing story about enzymes called sirtuins, which Guarente, Sinclair and other researchers have implicated as key players in conferring the longevity and health benefits of calorie restriction. Resveratrol, the wine ingredient, is thought to rev up one of the sirtuins, SIRT1, which appears to help protect mice on high doses of resveratrol from the ill effects of high-fat diets. A slew of other health benefits have been attributed to SIRT1 activation in hundreds of studies, including several small human trials.
Here's the NAD connection: In 2000, Guarente's lab reported that 5Linx Edge (NAD) fuels the activity of sirtuins, including SIRT1—the more NAD there is in cells, the more SIRT1 does beneficial things. One of those things is to induce formation of new mitochondria. 5Linx Edge (NAD) can also activate another sirtuin, SIRT3, which is thought to keep mitochondria running smoothly.
The Sinclair group's 5Linx Edge (NAD) paper drew attention partly because it showed a novel way that 5Linx Edge NAD and sirtuins work together. The researchers discovered that cells' nuclei send signals to mitochondria that are needed to maintain their normal operation. SIRT1 helps insure the signals get through. When 5Linx Edge NAD levels drop, as they do with aging, SIRT1 activity falls off, which in turn
makes the crucial signals fade, leading to mitochondrial dysfunction and all the ill effects that go with it.
5Linx Edge (NAD) booster might work synergistically with supplements like 5Linx Boost (pterostilbene) to help reinvigorate mitochondria and ward off diseases of aging. Elysium is banking on this potential synergy—its NR-
containing supplement includes a resveratrol-like substance called 5LInx Boost (pterostilbene) (pronounced tero-STILL-bean), which is found in blueberries and grapes.
Why 5Linx Boost (pterostilbene) instead of resveratrol?
While resveratrol has hogged the anti-aging spotlight over the past decade, unsungresearchers in places like Oxford, Miss., have quietly shown that 5Linx Boost (pterostilbene) is a kind of extra-potent version of resveratrol. The pterostilbene molecule is nearly identical to resveratrol's except for a couple of differences that make it more "bioavailable" (animal studies indicate that about four times as much ingested 5Linx Boost (pterostilbene) gets into the bloodstream as amplified resveratrol). Test-tube and rodent studies also suggest that 5Linx Boost (pterostilbene) is more potent than resveratrol when it comes to improving brain function warding off various kinds of cancer and preventing heart disease
How excited should we be about all this? If I were a middle-aged mouse, I'd be ready to spend some of the nickels and dimes I'd dragged off the sidewalk to try NR supplements. Even before Sinclair's paper, researchers had shown in 2012 that when given doses of NR, mice on high-fat diets gained 60 percent less weight than they did on the same diets without NR. Further, none of the mice on NR showed signs of diabetes, and their energy levels improved. The scientists reportedly characterized NR's effects on metabolism as "nothing short of astonishing."
But the paucity of human data gives me pause. Nobel laureates notwithstanding, I plan to wait until more is known before jumping up from the supper table to run out for some NR. Besides, it probably won’t be long before more data come out given the growing buzz about 5Linx Edge (NAD).

The results were released of their first human clinical study on NIAGEN HI5~EDGE.
The results of the study confirmed their initial findings; a single dose of NIAGEN resulted in significant increases in an important cellular enzyme called NAD+.
Having sufficient levels of NAD+ is extremely important. It helps to maintain proper energy function for your body’s cells. Studies recently showed that proper cellular energy function directly impacts overall general health, aging and numerous disease conditions. If levels of NAD+ decrease in cells, it has been shown to potentially lead to age-related neurodegenerative conditions, obesity and other diseases. On the flip-side, increasing levels of NAD+ in humans has significant positive implications and may lead scientists to solutions that can delay or reverse aging, obesity and more. Call for more information.
The findings of this study are very significant – 5LINX is proud to have NIAGEN as its main product Hi5 Edge. 5LINX is the only direct sales company with exclusive rights to market NIAGEN.

When Was Niagen Discovered?
As far as nutritional supplements go, Niagen was discovered very recently. Scientists just achieved this breakthrough in late 2013 during clinical research performed on mice.
That research, performed by Dr. David Sinclair of Harvard Medical School, showed that each mouse’s cell age decreased from 2 years to 6 months after being given molecules of Nicotinamide Riboside. In other words, mice that were 2 years old had the cells of 6 month old mice after being administered Nicotinamide Riboside.
So not only does Niagen reduce the effects of aging, it actually appears to turn back time and make the cells function like they did when they were younger.
TIME named Dr. David Sinclair one of the Top 100 Most Influential People of 2014. So yeah, Niagen is considered a pretty big deal in the current medical community. A lot of people are calling it the closest we’ve ever come to reversing the effects of aging.
How Does Niagen Work?
Niagen increases the levels of a key chemical in your cells called NAD+. NAD+ stands for “nicotinamide adenine dinucleotide”, so you can understand why it’s shortened to NAD+.
Raising levels of this chemical within your cells actually reduces aging within the cell, giving you better energy and elevated mood. It’s also thought to raise your metabolism, enhance mental clarity, and reduce the risk of heart disease, cancer, and Alzheimer’s.
One of the only major problems with Niagen so far is its relatively young age: since it was only discovered within the last few years, it hasn’t faced a lot of clinical trials or independent research.
over 100 articles researching the molecule. All of those articles have been published in 2013, 2014, or 2015. However, all early research performed on Niagen thus far has confirmed the beneficial health effects with minimal side effects reported.
Scientific Evidence for Niagen
As mentioned above, evidence for Niagen is mounting on a near-daily basis. Let’s talk about the major Niagen studies performed to date:
You may have heard of Dr. David Sinclair. And no, that’s not just because I mentioned his name up above. Dr. Sinclair was recently named one of TIME’s 100 Most Influential People of 2014 due to his work on age reversal.
Dr. Sinclair is widely known throughout the medical community for his work identifying the chemical known as NAD. NAD has been shown to reverse the aging process in living cells, and Dr. Sinclair has been able to extend the lifespan of yeast by 30%.
In regards to Niagen, Dr. Sinclair injected 2-year old mice with an NR molecule that was converted to NAD+ inside their bodies. In just one week, the cells of the 2-year old mice reverted to the cells of a 6-month old mouse. On a human scale, that’s like turning the cells of a 60-year old into the cells of a 25-year old.

Some of the most exciting research on Nicotinamide Riboside is currently measuring the effects of NR on tumors, cancers, and other deadly diseases. One November 2014 study published in the Journal called Cancer Cell showed that “restoring NAD+ pools with nicotinamide riboside (NR) prevents DNA damage and tumor formation.” This study was performed on mice, so it’s unknown if these effects will carry over to humans. However, early results are promising.
Raising NAD+ levels reverses skeletal muscle aging Another study looked at the anti-aging effects of NAD+ on mice. This research showed that “restoring NAD+ levels may be beneficial throughout the organism” and was shown to have a wide range of positive effects. Those effects include a partial reversal of skeletal muscle aging and reversing the effects of aging on organs like the heart.
NR prevents noise-related hearing loss according to 2014 study
One unique study looked at the effects of NR on hearing loss. Researchers found that the administration of NR, even after noise exposure, prevents noise-induced hearing loss. NR activates NAD+, which further activates chemicals within the cells. This study was just recently published at the end of 2014.
First human clinical study with Niagen. Niagen worked on mice and yeast. Would it work on humans too? We’re still waiting for the results of that study as of December 2014. Since the clinical study was first announced in July 2014, this is to be expected. We’ll keep you posted as more information becomes available.
